After entering a number of items in a purchase order, I had to go in and change an item (before posting). The part number was greyed out. I had to delete this line and then add a new line with the correct part. Is there a switch somewhere that allows line item editing and I've missed it? (Our old system allowed us to go in and change the part number).

The way we work with POA, the Item # is always greyed out as soon as we click OK to select the detail line.

I do not think that there is a switch that will give the option of changing an Item #, as this is a key field.
You are correct that you would have to delete the detail line and then insert the Item # that you want.

But maybe someone knows a trick that I am not aware of...

(You mention your "old system" - doesn't sound like Adagio.)
